The simple task of column and pH scouting can be easily automated using the ACQUITY UPLC^® System with the four-position ACQUITY UPLC Column Manager.

In this example, this configuration has been used to assess the selectivity of four different ACQUITY UPLC BEH chemistries (Phenyl C₁₈, RPC₁₈ and C₈) for the separation of the impurities of ranitidine HCl, a common histamine H2-receptor antagonist.

With a 5 to 95% methanol gradient at pH 9, the ACQUITY UPLC BEH Phenyl Column gave the best resolution of all of the peaks of interest.
