LC-MS Workflows for Confirmation, Characterization, and Impurity Profiling of Synthetic Peptides
Unparalleled Separation Performance
By combining our ACQUITY UPLC instrument technology with our industry leading Peptide Separation Columns, and our high quality MassPrep Peptide reference standard, Waters brings an unrivaled level of chromatographic performance to synthetic peptide analysis.
Fit-for-Purpose MS Detection & Informatics
As shown in the above graphic, our front-end separation technology can be paired with three “fit-for-purpose” mass detection and analysis options on the back-end to meet the needs of customers:
The ACQUITY QDa Detector workflow is our most deployable and scalable solution. It is ideal for customers looking to monitor peptide drug product quality/impurities profile. The added mass detection to the existing optical–based workflow provides an efficient and cost-effective option for improving confidence in synthetic peptide ID confirmation and purity assessments. This workflow can utilize either Empower CDS (Compliant-Ready Software) or MassLynx for instrument control and data acquisition. The MassLynx option is designed to work seamlessly with ProMass, or ProMass-HR, which enable automated spectral deconvolution and reporting of results.
The Xevo G2-XS QTof workflow delivers robust high-resolution mass spectrometry (HRMS) performance with isotopic resolution, and greatly enhanced sensitivity for in-depth characterization work, including MS/MS peptide sequence and impurities analysis. This workflow provides qualitative and quantitative identification and characterization of peptide drugs and related impurities with high selectivity, sensitivity, precision and good linearity. This workflow can utilize either MassLynx or UNIFI for instrument control and data acquisition. Data analysis and reporting for UNIFI acquisition uses UNIFI, and for MassLynx it is achieved through ProMass-HR.
The VION IMS QTof workflow delivers robust high-definition Ion Mobility mass spectrometry (HDMS) performance with isotopic resolution, greater selectivity and dynamic range, and greatly enhanced sensitivity for in-depth characterization work, including MS/MS peptide sequence and its related impurities confirmation. This workflow utilizes UNIFI Scientific Information System for instrument control, data acquisition, data analysis and reporting and provides qualitative and quantitative identification and characterization of peptide drugs and related impurities with high selectivity (as ion mobility cleans up and simplifies every spectrum), sensitivity, precision and good linearity.
Purification (Lab Scale)
Recognizing the need for fast and efficient purification at different scales and throughput, we have developed four efficient and reliable fit-for-purpose system solutions to address the needs of our customers. Operating at scales microgram to gram, from single samples to plates of samples, being purified in a high throughput environment, Waters Purification Solutions provide a tool to address all of your peptide purification needs. The use of MS triggered collection adds even greater specificity and efficiency to the purification workflow. By targeting only the component of interest, fewer unwanted components are collected. Waters Purification Solutions: AutoPurification Systems, ACQUITY UPLC H-Class Bio with Waters Fraction Manager-Analytical (WFM-A*), Prep 150 LC System, and Modular Systems. *Waters Fraction Manager Analytical (WFM-A) can also be paired with other Waters HPLC or UPLC systems.
