Optimized Reductive Amination Protocols

The GlycoWorks Sample Preparation consumable line along with the ACQUITY UPLC BEH Glycan Separation Technology Column is optimized for use with ACQUITY UPLC systems with fluorescence detection (FLR) for separating the released glycans of biopharmaceuticals as their 2-aminobenzamide (2-AB) derivatives.

The GlycoWorks Sample Preparation Consumable Line includes the following features:

Specifically for 2-AB labeled N-Glycans but the protocol can be adapted for 2-AA analysis
Works really well for the market of biopharmaceuticals that includes IgG’s (but not limited to this-just validated with IgG)

The HILIC Advantage

Hydrophilic Interaction Chromatography (HILIC) can be used to successfully improve retention of very polar species. This technique is rapidly growing for this application area. One reason for this is that this mode of separation is well suited for glycans due to their highly polar nature. The GlycoWorks HILIC SPE provides an efficient and reproducible mechanism for effective glycan cleanup and provides quantitative and consistent recoveries for a diverse range of N-Glycans.

Inclusive Methodology

The GlycoWorks Kit offers a protocol for complete sample processing of released N-glycans with either 2-AB or 2-AA labeling. Two formats offered, one for high-throughput which is a 96-well plate and the other for low volume users in a 1 cc cartridge format.

Sample cleanup devices with HILIC (amino-propyle) sorbent

High-throughput: GlycoWorks HILIC µElution SPE Plate (96-well)
