Waters patented RapiGest SF Surfactant was designed with your lab’s efficiency in mind, significantly decreasing proteolytic digestion time, and enabling same-day digestions with mass spectrometry (MS) compatibility.

RapiGest SF radically enhances the speed and percent recovery of protein enzymatic digestions. This patented anionic surfactant accelerates the in-solution production of peptides generated by proteases, such as Trypsin, Asp-N, Glu-C, Lys-C, and PNGase F. Many hydrophobic proteins are resistant to proteolysis because their cleavage sites are inaccessible to endoproteases. RapiGest SF facilitates the digestion of proteolytically resistant proteins and increases the solubility of hydrophobic proteins and peptides. RapiGest SF is an MS-compatible mild protein denaturant that helps solubilize and unfold proteins, making them more amenable to cleavage without denaturing or inhibiting common proteolytic enzymes like conventional denaturants. This enables use of lesser amounts of expensive endoproteases.

How RapiGest SF Surfactant works

RapiGest SF Surfactant is a mild denaturant that solubilizes and unfolds the substrate protein, making it more amenable to cleavage. Mild protein denaturation opens the protein structure and exposes the proteolytic sites to enzymatic cleavage. You can accomplish this during a more complex denaturation of globular proteins by heating in a RapiGest SF solution at elevated temperatures.


Compatible with MS

RapiGest SF is an acid labile surfactant that undergoes hydrolysis in acidic conditions. This unique feature allows you to utilize it to remove the surfactant from solutions when desired. Acidic conditions break down RapiGest SF to non-interfering byproducts, therefore peptide ionization is not suppressed.
