How RapiZyme works
RapiZyme Trypsin is a chemically optimized recombinant trypsin. Because it is not animal derived, it is free of contaminating chymotrypsin, does not require treatment with L-(tosylamido-2-phenyl) ethyl chloromethyl ketone (TPCK) inhibitors, and thus is free from non-specific digestion associated challenges.
Rapid Digestions
Extensive and homogenous methylation confers RapiZyme Trypsin with inordinately high autolysis resistance. This enables use of RapiZyme Trypsin at high E:P ratios (e.g., 1:5) to drive complete digestions in just 30-minutes, without resorting to higher temperature. Using Remicade® as a case study, RapiZyme Trypsin delivers <0.1% autolysis peaks and <1% missed cleavage (measured as % of total TIC response). An industry leading competitive trypsin delivers 6–7% autolysis peaks and 4–5% missed cleavage under the same conditions.
Clean baselines
Critical decisions are made by recognizing the smallest changes in a map. Regulatory agencies demand identification of every peak. Unmatched peaks, artifact peaks, autolysis peaks, and missed cleavage peaks are all time-consuming and unnecessary noises. They require extra scientific review and analysis, causing software processing times to shoot through the roof when non-tryptic peaks, or autolysis peaks from an in-silico model of trypsin digesting itself are included.

Reproducibility and purity – what’s in your trypsin?
RapiZyme Trypsin, homogenously methylated, recombinant (non-animal derived) trypsin is free of contaminating chymotrypsin. Further it is lyophilized with a LC-MS compatible excipient to make reconstitution easy and predictable and improve recovery from the vial.
The result is exceptional reproducibility both batch-to-batch and digestion-to-digestion.
Flexible to use
RapiZyme Trypsin can do everything you expect any trypsin (in solution or immobilized) to do and much more. With RapiZyme Trypsin, you can:
- Eliminate desalt & enhance consistency for MAM, PTM monitoring, bioprocessing and proteomics applications. RapiZyme trypsin maintains high activity with modest amounts of guanidine HCl
- Dilute guanidine HCl to 0.5–0.7M and digest in 2 hours at a 1:5 enzyme:protein ratio (pH 7.5)
- Control artifacts for any application when overnight digests are preferred without compromising digestion efficiency
- Digest overnight with a 1:100 enzyme:protein ratio at room temperature and pH 6.5
- Accelerate traditional 1:20 enzyme:protein ratio digests to 1–3 hours to afford additional flexibility
- Heat and eliminate the need for immobilized enzymes for simpler or easier to digest proteins with thermally stable RapiZyme
- Digest with a 1:10 enzyme:protein ratio for 1 hour at 70 °C
