RapiFluor-MS Label provides increased fluorescence quantification along with unprecedented mass spectral signal intensity. Easily assign and characterize low abundance glycans by serial fluorescence and mass spectrometric detection, using less sample while still obtaining increased sensitivity.
When labeled glycans were analyzed by HILIC chromatography, RapiFluor-MS Label produced the following results:
- Commercial rapid tagging analog of aminobenzamide (AB); 2x greater fluorescence signal and 1000x higher MS signal
- Conventional reductive amination with 2-AB; 4x greater fluorescence signal and 160x higher MS signal

Improved recoveries for disulfide rich complex molecules
N-glycans are routinely monitored for the quality of biotherapeutics because they affect the safety and efficacy of many therapeutic proteins. Current glycan analysis is developed with the well-studied glycans from monoclonal antibodies (mAb). There is a complementary method that provides a reproducible profile of N-glycans released from Human Chorionic Gonadotrophin (hCG), a highly complex glycoprotein with multiple disulfide bonds.

The loss of labeled acidic glycans – especially phosphorylated glycan species – during solid-phase extraction (SPE) purification is a challenge to accurately monitoring biotherapeutic glycosylation. The GlycoWorks Phosphoglycan SPE Elution Buffer, optimized with citrate additive, facilitates the elution, and recovery of phosphorylated glycans to achieve maximum yield.

The GlycoWorks Sample Preparation consumable line along with the ACQUITY UPLC BEH Glycan Column is optimized for use with ACQUITY UPLC Systems with fluorescence detection (FLR) for separating the released glycans of biopharmaceuticals as their 2-aminobenzamide (2-AB) derivatives.
The GlycoWorks Sample Preparation consumable line is specifically for 2-AB labeled N-glycans. This consumable line works especially well for the market of biopharmaceuticals, including Immunoglobulin G (IgG). The GlycoWorks Sample Preparation consumable line is not limited to IgG, but it has been validated with IgG.
