Intact protein separations with the Humanized mAb Mass Check Standard
Analyzing intact proteins by LC-MS is a preferred approach for biopharmaceutical characterization, especially monoclonal antibodies (mAbs). In mAbs analysis, it is possible to acquire intact mass information, quickly confirm sequence and post-translation modifications, and interrogate product-related impurities. To support this application area, Waters designed the Humanized mAb Mass Check Standard, which can be used to benchmark LC-MS techniques, proficiency test among different instruments and laboratories, and ensure system suitability for intact protein analysis. This protein standard is a stabilized and lyophilized format of NIST Reference Material 8671 (NIST mAb), a humanized IgG1κ expressed from a murine cell line.*
Subunit analysis with the mAb Subunit Standard
Waters mAb Subunit Standard can be used in the benchmarking of LC and LC-MS techniques, proficiency testing among different instruments and laboratories, and system suitability. This standard is filtered and stabilized formulation of reduced, IdeS-digested NIST Reference Material 8671 (NIST mAb), a humanized IgG1к expressed from a murine cell line. Each vial of the standard is provided in an approximately 25 µg quantity.
Size-exclusion separations with the SEC Protein Standard Mixes
The SEC Protein Standard Mixes are unique, specially formulated protein mixtures specific to the ACQUITY UPLC BEH 200Å, BEH 125Å, and BEH 450Å Columns. Using SEC Protein Standard Mixes regularly is recommended to evaluate these types of columns for optimal system performance.
IEX with the Charge Variant Standard
Waters mAb Charge Variant Standard can be used in the benchmarking of LC and LC-MS techniques, proficiency testing among different instruments and laboratories, and system suitability. This standard is intended to be used with BioResolve SCX mAb, 3 µm Columns and BioResolve CX pH Concentrates. It is formulated as filtered and stabilized formulation of a void marker (tryptophan), conalbumin from chicken egg white, and NIST Reference Material 8671 (NIST mAb), a humanized IgG1к expressed from a murine cell line. Every standard vial contains approximately 0.5 µg of tryptophan, 200 µg conalbumin, and 100 µg of NIST mAb.
Hydrophobic interaction chromatography with HIC Protein Standard Mix
HIC Protein Standard Mix is used to benchmark HIC Columns, especially the Waters Protein-Pak Hi Res HIC Columns. Using this standard regularly is recommended to evaluate column and system performance and ensure confidence in results. This protein standard is uniquely formulated and optimized with a 6-component intact protein mixture to span a typical chromatographic range in this application.
